The incoming material

Precursor processing

A delivery can be transformed on its way. The precursor research gives the incoming side of the account a surprisingly indirect journey.

Making NAD+ from precursors

NMN is nicotinamide mononucleotide; NR is nicotinamide riboside. They enter discussions of NAD+ because they are materials from which it can be made. In structural work on human NR kinase, with supporting yeast experiments, the enzyme’s shape helped explain how NR is phosphorylated along a route to NAD+.

Phosphorylation means adding a phosphate group. The appeal of this paper is its physical specificity: an enzyme, a substrate and a reaction, rather than a vague impression that a named ingredient arrives intact wherever it is wanted. I like being able to follow an actual conversion.

The broader review of NAD+ homeostasis describes several ways a supply can become inadequate: impaired production, increased consumption or dietary deficiency of vitamin precursors. That variety makes an incoming material only one part of the account.

A mouse isotope-tracing experiment

In isotope-tracing experiments in mice, circulating host nicotinamide fed the gut microbiome. Microbes converted it to nicotinic acid that supported NAD+ synthesis in host tissues. Oral NR reached host NAD+ mainly through microbial conversion to nicotinic acid.

An isotope tracer lets investigators follow material through transformations. The circuit is the intriguing part: the host supplies microbes, and microbial processing returns material to the host. My tidy mental picture of a delivery has acquired a workshop along the route.

This is mouse metabolism. A human preference between precursors remains unresolved here, and the experiment supplies no reason to improvise a human regimen. It does show why the name on the incoming material may be an incomplete description of its journey.

Replenishment in human studies

The human precursor review reports consistent increases in blood NAD+, limited clinical efficacy and sparse tissue-specific information. Those are separate entries in the account: delivery, destination and consequence.

I’d resist crowning an ingredient on the strength of the delivery alone. The muscle essay offers concrete examples of consequences that were measured, including insulin sensitivity and walking distance. The consumption essay returns to the other side of the ledger: the jobs drawing on the supply.
